ИЗМЕНЕНИЕ ЭКСПРЕССИИ ИЗОФОРМ МИКРОРНК И ИХ МИШЕНЕЙ ПРИ ГИПОКСИИ В КЛЕТКАХ HT-29
Tumor hypoxia is one of the main reasons of progression and metastasis of colorectal cancer. The important molecular mechanism of cell response to hypoxia consists in up- or down-regulation of microRNA expression. In this work we performed microRNA and mRNA sequencing of human colorectal adenocarcinoma HT-29 cell lines treated with two chemical agents mimicking hypoxia: cobalt (II) chloride and oxyquinoline. Bioinformatics analysis of the obtained data revealed differentially expressed isomiRs (hsa-miR-210-3p|0, hsa-miR-22-3p|0, hsa-let-7a-3p|0, hsa-miR-615-3p|0, hsa-miR-4521|0) and their targets, which changed their expression upon hypoxia induction by both treatments. Thus, we identified new regulatory mechanisms of cellular response to hypoxia.
