Methionine gamma lyase from Clostridium sporogenes increases the anticancer effect of doxorubicin in A549 cancer cells in vitro and human cancer xenografts
Ch. 18. P. 243–261.
Манухов И. В.
The anti-cancer efficacy of methionine γ-lyase (MGL) from Clostridium sporogenes (C.
sporogenes) is described. MGL was active against cancer cells in vitro and in vivo. Doxorubicin
(DOX) and MGL were more effective on A549 human lung cancer growth inhibition than either
agent alone in vitro and in vivo. The growth inhibitory effect of DOX + MGL on A549 xenograft
in vivo was reflective of the results obtained in vitro.

The principal possibility of inhibition of histone deacetylases (HDACs) in nuclear extract of HeLa cells by N1-hydroxy-N4-(pyridine-4-yl)succinamide (compound 1) has been investigated. Compound 1 inhibits HDAC and exhibits low toxicity to A-172, HepG2, HeLa, MCF-7, and Vero cells. HeLa cells are most sensitive to the compound. With an increase in the interval between the administration ...

New platinum(IV)-aminoxyl complexes (PAC) 2a,b have been synthesized, containing aminoxyl radical with antioxidant properties in one of equatorial position, and axial ligands are represented by dichloroacetate, capable of inhibiting energy production through glycolysis in tumor cells. Complexes 2a,b are characterized by moderate lipophilicity (lg Pow ~2) and differ in the redox properties of aminoxyls. Cytotoxic ...

Compositions of doxorubicin with a terpolymer of N-vinylpyrrolidone, methacrylic acid and triethylene glycol dimethacrylate have been prepared and investigated by UV-VIS and NMR spectroscopy, cyclic voltammetry and DFT quantum chemical modeling. Oxygen containing groups of the terpolymer and hydrogen atoms of the drug OH and NH3+ groups form hydrogen bonds. The compositions have lower cytotoxicity in ...

Nanoplatforms applied for the loading of anticancer drugs is a cutting-edge approach for drug delivery to tumors and reduction of toxic effects on healthy cells. In this study, we describe the synthesis and compare the sorption properties of four types of potential doxorubicin-carriers, in which iron oxide nanoparticles (IONs) are functionalized with cationic (polyethylenimine, PEI), ...

Objectives: The study of the transport and toxicity of xenobiotics in women is limited for ethical reasons. Ex vivo placenta models have high variability and low success rates. Animal models in vivo differ from a human in anatomy, genotype, and proteome. The placenta-on-a-chip model is a compromise. We studied the components of the FAC chemotherapy regimen for breast cancer in ...

An in vitro placental barrier model based on human choriocarcinoma BeWo b30 cell line was considered as a method of preclinical study of the transport and toxicity of antitumor agents and other organic compounds. Low permeabilities were found for 5-fluorouracil as an example of hydrophilic compound and for doxorubicin as an example of a lipophilic compound with ...

Cancer cells require exogenous methionine for survival and therefore methionine restriction is a promising
avenue for treatment. The basis for methionine dependence in cancer cells is still not entirely clear. While
the lack of the methionine salvage enzyme methylthioadenosine phosphorylase (MTAP) is associated with
methionine auxotrophy in cancer cells, there are other causes for tumors to require exogenous ...

Preparation of liposomal formulations containing water-soluble drugs in the form of lipophilic
prodrugs in their lipid bilayer is of considerable interest. Previously, we synthesized doxorubicin dioleoyl
glyceride and oleoyl conjugates intended for incorporation into fluid-phase liposomal bilayers. In this work,
we studied the behavior of lipid conjugates in bilayers prepared from palmitoyl oleoyl phosphatidylcholine
and dimyristoyl phosphatidylcholine using methods ...

We studied the expression of peroxiredoxin genes (PRDX1, PRDX2, PRDX3, and PRDX6) in human erythroleukemia K652, human breast carcinoma MCF-7, and human ovarian carcinoma SKOV-3 cells during cisplatin resistance development. It was found that drug resistance formation was accompanied by a significant increase in the expression of PRDX1, PRDX2, PRDX3, PRDX6 genes in all cancer ...
